PPROBLEM TO BE SOLVED: To provide a method for directly, simply and easily synthesizing a luminescent substrate of luminescent coleopteran luciferase useful as a reporter gene for detecting the gene expression of a mammal from natural L-cysteine having a wide application range as a base in a reaction solution, and to provide a method for directly synthesizing the luminescent substrate of firefly luciferase from the natural L-cysteine as a base in a reaction solution, in a method for measuring cysteine, ATP and gene expression. PSOLUTION: This method for producing D-firefly luciferine or its luminescent derivative comprises reacting a L-firefly luciferine raw material comprising at least one selected from the group consisting of (1) the ester of L-firefly luciferine or its derivative and (2) the thioester of L-firefly luciferine or its derivative with an esterase.
