PPROBLEM TO BE SOLVED: To provide a method for examining the diathesis of insulin resistance or type 2 diabetes by detecting AMPK gene polymorphism. PSOLUTION: This new SNP (single nucleotide polymorphism) of PRKAA2 gene is found out by comparing the gene sequences of DNAs derived from a type 2 diabetes patients with a database, GenBank. By performing a linkage disequilibrium analysis on 10 pieces of SNPs including the new SNP, as a result, the 10 pieces of SNPs are classified into 6 clusters. By analyzing the relationship of haplotypes prepared by selecting one each SNP from each of the clusters, with the diabetes, it is found out that the haplotype having a minor (A) allele regarding to an SNP: rs2051040, and the haplotypes having major alleles regarding to the other 5 SNPs are significantly related with the diabetes, and also by examining the relationship of the above 10 SNPs with the insulin resistance, it is found out that the rs2051040 is related to the insulin resistance.
