GENE NYC1 PARTICIPATING IN DEGRADATION OF CHLOROPHYLL AND ITS UTILIZATION

PPROBLEM TO BE SOLVED: To relate to a gene participating the degradation of chlorophyll and to the green color maintenance of a plant by the utilization of the gene, and to control the chlorophyll degradation (etiolation) of the plant to enhance an ability for maintaining the green color. PSOLUTION: The isolation of a causative gene from a mutant nyc1-1 yellowing the leaves even on ageing by a positional cloning was tried, and it was consequently found that an etiolation-related gene NYC1 existed between P0452F10.5 gene and P0443E07.24 gene. When the base sequence of a region between the two genes in the mutant nyc1-1 was analyzed, the presence of one base replacement of P0452F10.4 gene existing between both the genes was confirmed, and it was estimated that the abnormal splicing of the gene was caused in the mutant nyc1-1. Additionally, when the above-mentioned region in a cultured mutant nyc1-2 exhibiting a green color phenotype was analyzed, it was found that the P0452F10.4 gene was degraded with a transposon. When a genome fragment containing wild type NYC1 was introduced into the green color-maintaining mutant nyc1-2, it was confirmed to show etiolation similarly to a wild type. An ability for enhancing the green color can be enhanced by controlling the function of the etiolation-related gene NYC1. PCOPYRIGHT: (C)2006,JPO&NCIPI
