PPROBLEM TO BE SOLVED: To provide a composition capable of restraining the diazonium coupling of the background and a color change thereof to a minimum, when measuring an analyte selected from among leukocyte, esterase and protease in a sample, and to provide a testing tool and a testing method that uses the same. PSOLUTION: The composition for measuring the presence of the analyte is a composition containing diazonium salt, ester hydrolyzed in the presence of the leukocyte, esterase or protease, and salt of an alkaline earth metal, and the testing tool and a testing method using the same are provided also.
