Methods and compositions for treating inflammatory and proliferative diseases,enabling treatment of MDR tumor cells by using combination of compounds, or abifunctional compound, that inhibits both fatty acid metabolism andglycolysis. In particular, the invention combines or links a glycolysisinhibitor that is, or that is derived from, hypoglycin A (also referred to ashypoglycine A) and a fatty acid metabolism inhibitor. Preferably, theinvention provides bifunctional compounds that link a moiety having thefunctionality of an oxirane carboxylic acid compound to a moiety having thefunctionality a hypoglycin A derivative. In specific embodiments, theinvention provides a bifunctional compound that links a moiety having thefunctionality of etomoxir to a moiety having the functionality of hypoglycin A.
