It is intended to provide a method of imparting to cells of a microorganism or the like a tolerance against short-chain fatty acids such as formic acid and acetic acid, which are harmful to the growth thereof, without inhibiting the growth of the cells while sustaining a high productivity of a useful substance; a means of efficiently culturing a microorganism in the presence of short-chain fatty acids; and a means of producing a useful short-chain fatty acid via fermentation. Namely, a method of breeding cells having an improved tolerance to short-chain fatty acids characterized by comprising transferring a gene encoding a protein, which has a function of discharging outside short-chain fatty acids from cells, into the cells and expressing the gene; a high-density cell culture method characterized by comprising using the cells as described above; and a method of producing a fermentation liquor containing a short-chain fatty acid characterized by comprising culturing the above-described cells under such conditions as allowing the formation of the short-chain fatty acid by the cells.
