Detecting agonists of insect calcium ion channels, useful potentially as insecticides, by measuring their effect on recombinant cells that express the channel and calcium-sensitive luciferase-binding protein
Detecting an agent (I) that activates an insect calcium ion channel (A) regulated by cyclic nucleotides, is new. Detecting an agent (I) that activates an insect calcium ion channel (A) regulated by cyclic nucleotides comprises: (a) contacting a recombinant eukaryotic cell (B), genetically modified to express insect CNG-1 (cyclic nucleotide gated-1 channel) and a calcium-sensitive luciferase-binding apoprotein (AP), with a coelenterate luciferin, so that includes a calcium-sensitive luminescent holoprotein (hP); (b) contacting the cells with a test agent and a calcium-chelating agent, to produce a test sample; (c) treating the sample with an excess of calcium ions; and (d) determining any effect of the test agent on CNG-1 activity, by measuring the level of intracellular calcium, where an increase in this level indicates a CNG-1 agonist.
