PPROBLEM TO BE SOLVED: To provide a method for producing a non-natural-type amino acid-incorporated protein in eukaryocyte using an aminoacyl tRNA synthase and an aminoacyl tRNA, to provide a method for producing such an aminoacyl tRNA in eukaryocyte, and further, to provide a tRNA, and to provide a nucleic acid to be used for synthesizing such a tRNA. PSOLUTION: The method for producing a non-natural-type amino acid-incorporated protein comprises expressing an aminoacyl tRNA synthase, a nucleic acid comprising a sequence obtained by binding a eukaryote-derived tRNA nucleic acid sequence to the 5'-terminal of the nucleic acid sequence of a tRNA bindable to a non-natural-type amino acid in the presence of the aminoacyl tRNA synthase, the non-natural-type amino acid and a desired protein gene subjected to nonsense mutation at a desired position, and incorporating the non-natural-type amino acid in the position of the nonsense mutation of a protein to express the objective non-natural-type amino acid-incorporated protein.
