PROBLEM TO BE SOLVED: To provide a method with which the amounts of cAMP and cGMP in a single biological sample of a cell or a tissue are measured simultaneously with high sensitivity.;SOLUTION: The measuring method comprises the following steps: the GMP AMP ADP and ATP in a biological sample are decomposed by using an apyrase, an alkaline phosphatase and an adenosine deaminase; subsequently, the GDP in the sample is converted into GTP by using a creatine kinase and creatine-1 phosphoric acid; after the GTP in the reaction mixture is adsorbed by a column, the cAMP and cGMP in the reaction mixture are selectively eluted; the cAMP and cGMP are converted into AMP and GMP respectively by using a cyclic nucleotide selective phosphodiesterase, then converted into ADP and GDP, and successively to ATP and GTP by an enzyme-multiplied reaction; the pyruvic acid formed during these reactions is converted into 2- hydroxynicotinaldehyde; and the fluorescence intensity of the 2- hydroxynicotinaldehyde is determined.;COPYRIGHT: (C)2003,JPO
