The method involves realizing an enrichment of an acid function present in an oil sample e.g. crude oil, using an isotope ratio mass spectrometry, and determining an enrichment value of the sample. An acidity of the sample is deduced form the enrichment value. The enrichment value determination step involves measuring isotopes of a chemical element of the sample before and after the enrichment.
