A cocktail kit for high-throughput evaluation of human cytochrome P450 enzyme is provided to inhibit interaction with 5 probe drugs and side effects, and increase evaluation speed. The cocktail kit for high-throughput evaluation of human cytochrome P450 enzyme comprises probe drugs consisting of caffeine, losartan, omeprazol, dextromethorphan and midazolam, wherein the human cytochrome P450 enzyme is selected from CYP1A2, CYP2C9, CYP2C19, CYP2D6 and CYP3A; the probe drugs are orally administered simultaneously or sequentially; and the amounts of caffeine, losartan, omeprazol, dextromethorphan and midazolam are 50-200 mg, 25-200 mg, 10-40 mg, 10-50 mg and 1-5 mg, respectively.
