An apparatus and a method for selecting an RS(Relay Station) by using an RS preamble signal in a multi-hop relay BWA(Broadband Wireless Access) communication system are provided to enable MSs(Mobile Stations), which directly communicate with BSs(Base Stations) to use relay communication capable of increasing effective transfer rates through excellent radio link channels, provided by RSs(Relay Stations), as a BS allocates a channel domain to an RS in order to grasp the channel state information between an RS and an MS and receives an RS-MS signaling level measurement value from the MS. A BS transmits preamble channel information to RSs(421). Each of the RSs transmits a preamble to each MS by using the preamble channel information(425). Receiving the preamble, each MS measures the signaling level of each RS and reports a measured result to the BS(427,429). The BS selects a suitable RS for each MS based on the reported results(431).
