A method for producing YMV-K free yam plants by the cryopreserving and regenerating of cultivated yam and wild yam is provided to effectively preserve the yam plants without genetic mutation for a long period of time, and remove YMV-K virus from the yam plants without additional treatment, so that the YMV-K free yam plants are mass produced stably. The method for cryopreserving and regenerating the cultivated yam seeds comprises the steps of: (a) pre-culturing the shoots of cultivated yam in a medium containing sugar; (b) capsulating the pre-cultured shoots with Na-alginate solution; (c) drying the shoot capsules under antiseptic condition, and preserving them in liquid nitrogen; (d) dissolving the cryopreserved shoot capsules; and (e) culturing the dissolved shoot capsules in a medium containing BAP or a mixture of BAP and kinetin as plant growth regulators, wherein the sugar in step(a) is 0.3M sucrose or sorbitol. The method for producing YMV-K free cultivated yam comprises the steps of: (a) culturing the nodal pieces of cultivated yam seeds; (b) inducing in vitro microtubers of the cultivated yam seeds in a medium containing GA3; (c) preserving the in vitro microtubers at 4 deg.C for 2 months or more; and (d) sprouting the preserved in vitro microtubers of cultivated yam.
