A method for determining the concentration of homocysteine in plasma by using gas chromatography-mass spectrometer is provided to prevent re-oxidation of homocysteine reduced in a pretreatment step, thereby permitting accurate determination, and to contribute to studies about a heart disease, stroke, dementia, Alzheimer's disease. A method for determining the concentration of homocysteine in plasma by using gas chromatography-mass spectrometer(GC-MS) comprises the steps of: removing homocysteine-bonded protein; reducing homocysteine; non-polarizing homocysteine; and determining the concentration of homocysteine in plasma by using GC-MS, wherein the step of removing homocysteine-bonded protein is carried out by using acetone at 3-5 deg.C.
