Designing oligonucleotides used in molecular biology process comprises selecting/creating a data base of reference sequences, i.e. an evaluating data base, selecting a subset of sequences of a desired organism to create a reference data base, i.e. a design data base, selecting oligonucleotides to be examined from the subset of sequences, purifying the selected oligonucleotides as per the criteria of hybridization specificity and thermodynamic stability and obtaining a list of oligonucleotides that satisfies all the above conditions to give oligonucleotides obtained by the method.
