PPROBLEM TO BE SOLVED: To provide a high-efficient and convenient method for synthesizing a phosphorylated protein. PSOLUTION: The method for synthesizing a phosphorylated protein of the present invention comprises the following steps of: synthesizing a modified tRNA in which the anticodon of cysteine tRNA is substituted by a four base anticodon consisting of either one base sequence selected from AGCA. CGCA. GGCA. UGCA, GCAG, GCAU, GCAC and GCAA in a cysteine tRNA derived from archaebacteria having synthesizing ability for Sep-tRNA; aminoacylating Sep with the modified tRNA using SepRS; introducing the base sequence of a codon which corresponds to the four base anticodon contained in the modified tRNA into the site in which Sep is introduced in DNA encoding the target protein; obtaining mRNA derived from the modified DNA; and synthesizing a protein using a translation system comprising Sep-tRNA and mRNA.
