PPROBLEM TO BE SOLVED: To provide a primer set enabling the amplification of a nucleic acid in a high amplification efficiency and in high sequence specificity. PSOLUTION: This primer set comprising at least two primers amplifying a target nucleic acid sequence is characterized in that at least one primer contained in the primer set (i) can hybridize with the 5'-terminal portion of a primer extended strand on the identical strand in the primer extended strand obtained by the extension reaction of the primer on a template and (2) can provide an endonuclease-recognizing site, when the sequence of the primer becomes a double strand state by the synthesis of a complementary strand by a nucleic acid amplification reaction, and the endonuclease exhibits nicking activity.
