PPROBLEM TO BE SOLVED: To provide a method for evaluating the proliferation-participating property of DNA in a bacterium belonging to Bacilli class, and to provide a method for identifying a proliferation-participating region. PSOLUTION: This method for evaluating the DNA proliferation-participating property of the DNA comprises forming a low copy plasmid having a par gene participating in the stable holding of a plasmid, a replication initiator, and a rep gene to form the plasmid stably existing in bacteria belonging to Bacilli class, such as Staphylococcus aureus, and then utilizing a plasmid shuffling with the formed plasmid.
