E. BSMBI restriction endonuclease and methylase BSMBI cloning and methods of expression in COLI, and BSMBI endonuclease purification methods

PROBLEM TO BE SOLVED: To provide a recombinant DNA encoding BsmBI restriction endonuclease as well as BsmBI methyltransferase and expression of the BsmBI restriction endonuclease and BsmBI methylase in E. coli cells containing the recombinant DNA, and further a method for purifying the recombinant BsmBI restriction endonuclease.;SOLUTION: The isolated recombinant DNA encodes the BsmBI restriction endonuclease and is available from Bacillus stearothermophilus B61. The method for cloning and expression of the BsmBI restriction endonuclease of the Bacillus stearothermophilus B61 in the E. coli by methylase selection and inverse PCR is provided.;COPYRIGHT: (C)2003,JPO
