the present invention relates to methods for the regeneration and transformation of durum wheat varieties adapted to the conditions in africa and the middle east. methods are described for the production of callus, somatic embryos and plants of wheat from mature and immature embryos.four varieties of wheat cultivated in tunisia were first tested for their ability to regenerate from immature embryos and explants used in the in vitro culture in wheat. the results showed a highly genotype dependent.one variety is noted for its percentage of callus induction, callus, embyogu00e8nes and the average number of plants per explant in culture. this variety has been the potential devaluation of regeneration from mature embryos.the results showed that the variety has a potential for regeneration from mature embryos is approximately 5 times higher than that from immature embryos. agrobacterium transformation of mature embryos of this variety has resulted in the stable integration of the transgene with a processing rate of about 9%.the invention is useful because it allows the use of mature embryos in the process of the production of durum wheat varieties.accelerating technological advances related to the field of commercial varieties of durum wheat and eliminates the use of on the immature embryo explants and other types of the production is still a difficult and not available throughout the this year.
