Disclosed is a method for detecting a nucleic acid target sequence ofinterest. Themethod involves the addition of probes complementary to regions of a double-strandedpromoter, to form a complex comprising three strands of nucleic acid, suchthat afunctional promoter may be formed within the complex suitable to receive apolymerasethat recognizes the formed promoter. De novo synthesis via the polymerase fromtheformed promoter allows for the production and subsequent detection of asynthesizednucleic acid. Also disclosed is the complex formed in performance of themethod, and akit for performing the method.
