METHOD OF PRODUCING SMALLPOX VACCINE USING CELL CULTURE AND THE SMALLPOX VACCINE PRODUCED BY THE METHOD

A method for producing a smallpox vaccine is provided to improve use safety and quality of smallpox vaccine, and inhibit weakness of immunogenicity and degeneration during the long-term storage of the vaccine by using cell culture. A method for producing a smallpox vaccine comprises the steps of: inoculating 0.001-0.1 M.O.I.(multiplicity of infection) of attenuated vaccinia virus NYCBOH(New York city board of health) strain derived from the CJ-MVB-SPX master virus bank into human derived MRC-5 cell line, and culturing the infected cell line in serum free medium for 46-48 hours; and freezing, melting and sonicating the recovered cells from the cultured medium to release attenuated vaccinia virus CJ-50300(KCCM 10797), and recovering it. An immunogenic composition for preventing and treating smallpox comprises the attenuated vaccinia virus CJ-50300(KCCM 10797) and a stabilizer containing NaCl, KCl, Na2HPO4.12H2O, KH2PO4, sucrose, sodium L-glutamate, gelatin, hydrolyzed gelatin, EDTA(ethylene diamine tetraacetic acid) trisodium salt and distilled water.
