PROBLEM TO BE SOLVED: To provide a new, simple, and economically superior fluorescence staining method of a biological specimen to be tested and to provide a kit for assay used for the method.;SOLUTION: This method for discriminating the presence of a target analysis substance in the biological specimen to be tested by detecting a measurable signal includes a process (a) for making a first substrate, formed by immobilizing the biological specimen to be tested onto a solid-phase matrix, come into close contact with a second substrate, having two partitions or more including a specifically binding substance to the target substance, in such a way that the biological specimen to be tested is in contact with the specifically binding substance; a process (b) for performing incubation on conditions sufficient for completing the binding reaction between the target analysis substance and the specific binding substance in the case that the binding reaction occurs; and a process (c) for verifying the measurable signal in the biological specimen to be tested after the incubation.
