PROBLEM TO BE SOLVED: To provide a method for cloning a PCR product obtained by using a DNA polymerase having 3'-5' exonuclease activities and having a base sequence with high accuracy without purifying the product by incorporating the product into a vector.;SOLUTION: A reaction for inhibiting the activity of the DNA polymerase having the 3'-5' exonuclease activities and a reaction for adding dA to the 3' terminal of the PCR product are carried out in one container in a pretreatment step of the cloning. By using the method, the PCR product amplified by a highly accurate method can be simply cloned.;COPYRIGHT: (C)2004,JPO
