The present invention provides methods of identifying the presence of a target nucleic acid in a sample wherein the target nucleic acid is replicated and said replication of the target is detected as the consumption of a deoxynucleotide triphosphate precursors wherby any release of PPi is indicative of incorporation of deoxynucleotide or dideoxynucleotide and thus replication of the target DNA. The invention also provides kits and reaction mixtures utilized in the methods of the invention.
