Provided is a high-density fermentation method of Chlorella protothecoides by heterotrophic culture in bioreactor for biodiesel production, which comprises the steps of: selecting a strain with fast growth rate and high lipid content, primary seed culture in shaking flasks, secondary high-density fermentation with condition optimization and process control, collecting and freeze-drying Chlorella cells, extracting total lipid and preparing biodiesel by transeterfication. The method reduces the material cost of biodiesel production and meets the requirement of industrial application of heterotrophic culture of Chlorella protothecoides for biodiesel production.
