A method for preparing doughnut-shaped cell capsule is provided to improve cell viability and function by improving supply of nutrient and oxygen to the cell, so that the capsule is useful for cell therapy. The method for preparing doughnut-shaped cell capsule comprises the steps of: mixing cells with protein-based hydrogel and biodegradable polymer; and forming the doughnut-shaped flat cell capsule in which the center has a hole by placing the mixture under acidic condition for 2-3 hours through self assembly.
