An expression cassette, the chimeric plasmid and recombining vector are provided, which express the selenoprotein M. A manufacturing method of cassette comprises: a step for amplifying human selenoprotein base sequence by using PCR; a step for cloning to the pGEM T vector; and a step for making the pCMV/EGFPhSelM vector by combining with the pEGFP C-1 vector after separating the phSelM-T vector by using the SalI limitation enzyme.
