Recombinant gene for mass production of modified tissue type plasminogen activator and protein refolding method thereof

A modified recombinant tissue type plasminogen activator(tPA) is provided to show similar activity to that of a natural tPA derived from a CHO cell, maintain the biological activity and be used as an economical thrombolytic therapeutic agent. A tPA recombinant protein is described in a sequence listing 2 where a codon is substituted in a sequence of an amino terminal of a protein of the tPA. An expression vector pM2tPA includes a plasminogen activator recombinant gene described in a sequence listing 1 encoding the recombinant protein. A bacteria cell is transformed by the expression vector. A method for preparing the tPA protein comprises the steps of: (a) mass-expressing a recombinant tPA inclusion body in the transformed cell; (b) isolating the recombinant tPA inclusion body expressed in the cell; (c) treating a protein dissolution solution with the isolated protein inclusion body; and (d) treating a re-folding reaction mixture with the recombinant protein dissolved in the dissolution solution to progress a refolding condition.
