A quantitative magnetic cell sorting method using a second spectral image system is provided to count the object automatically by obtaining the equivalent dispersion of the threshold parameter about all objects. A quantitative magnetic cell sorting method using a second spectral image system comprises a platform placing a sample; a white light source employing the ion laser light source or the filter; an ND(Neutral Density) filter reducing the incidence quantity of the incident light; an interference filter refining the incident light passing through the neutral density filter; a dichromatic mirror passing the collected fluorescent beam through from the objective lens; an objective lens collecting the excited fluorescent beam from sample; a prism reflecting the fluorescent beam; lens reducing the diameter of the fluorescent beam passing through prism; a variable form filter controlling the wavelength of the fluorescent beam passing through the lens according to the time; a long- pass filter which prevents to scatter of beam; a CCD(Charge Coupled Device) camera detecting the fluorescent beam passing through the long-path filter; and a computer system equipped with the image captured and analysis software of the fluorescent beam detected by the CCD camera.
