PPROBLEM TO BE SOLVED: To simultaneously progress bare nuclearization using a surface-active agent, the removal of RNA, and the dyeing of fluorescent light by substantially simultaneously adding them into a cell suspension liquid. PSOLUTION: A method of measuring the amount of DNA in a cell nucleus includes: a dyeing process for substantially simultaneously adding the surface-active agent, an RNA removal liquid, and a fluorescent dyeing coloring matter to the cell suspension liquid for reaction for a prescribed amount of time; and an analysis process for measuring the amount of cell nucleus DNA in the cell suspension liquid dyed by the dyeing process using a flow cytometer.
