PROBLEM TO BE SOLVED: To provide a fertility diagnostic method for estimating fertility on the basis of fusion ability of sperm to ovum and identifying cause of infertility, a polynucleotide and a fertility diagnostic kit used for the method.;SOLUTION: The fertility diagnostic method includes a detection step for using a biological sample collected from human and detecting presence of deficiency or mutation of Calr3 gene in the human. In the polynucleotide comprising a base sequence of human wild type Calr3 gene or a base sequence of translated region of the gene when adenine of translation initiation codon is a first base, the polynucleotide comprises 10-100 continuous base sequences containing a mutation site of single base substitution mutation having high correlation to fertility.
