The present invention relates to a method of diagnosing cancer using dosage-sensitive genes through the degree of methylation of the transitional zone of the genes. The invention provides a biomarker comprising a dosage-sensitive gene(s), which lacks CpG islands, is rich in Ll elements, has low BOE (breadth of expression) and high PHE (peak height of expression) and contains a transitional zone defined as a chromosomal region between the transcription start site of the gene and its nearest retroelement. The invention also provides a primer which binds complementarily to the biomarker. The biomarker and the primer can be advantageously used to predict cancer development and progression and a predisposition to cancer in normal persons.
