PURPOSE: A substrate of neuron chip for improving survival rate of nerve cells is provided to enable patterning without coating cell adhesion material. CONSTITUTION: A method for preparing a substrate of neuron chip comprises: a step of mixing 10-20 weight parts of hardener based on 100 weight parts of polydimethylsiloxane(PDMS) to mold PDMS substrate; a step of treating the surface of PDMS substrate with O2 plasma to make in hydrophilic surface; a step of sterilizing the PDMS substrate; a step of coating the surface with cell adhesion material; and a step of culturing the nerve cells.
