PURPOSE: A device for separating a target material and a method of purifying the target material are provided to collect a target material using a microparticle and to separate the microparticle from the fluid comprising the target material.;CONSTITUTION: A device(1) for separating a target material comprises a filter unit(200), a fluid providing part(100), and a resupply unit. The filter unit comprises an inlet, a separating unit, an outlet and a filter, which blocks the leakage of the microparticle through the outlet. The fluid providing part selectively provides a sample solution and eluent to the filter unit. The resupply unit resupplies the eluent passing, through the filter unit, to the filter unit. The resupply unit comprises a resupply path(510) and a resupply pump(520). The resupply path connects the outlet to the inlet.
