The isolation method of monocytes from peripheral blood mononuclear cells by flotation density gradient centrifugation

A method for isolating monocyte from peripheral blood mononuclear cell (PBMC) with floating density gradient centrifugation is provided to raise the purity and yield of monocyte by regulating density and osmotic pressure of histopaque or ficoll solution. A method for isolating monocyte from peripheral blood mononuclear cell (PBMC) with floating density gradient centrifugation is centrifuging PBMC using histopaque or ficoll solution. A density and osmotic pressure of histopaque or ficoll solution are 1.071-1.073g/ml and 330-340mOsm, respectively. A method for applying the density and osmotic pressure of histopaque or ficoll with determined figure is mixing PBS and NaCl solution in histopaque or ficoll solution. A dendritic cell is obtained by culturing the isolated monocyte.
