PPROBLEM TO BE SOLVED: To provide a composition and a method for enhanced in vitro synthesis of polypeptides. PSOLUTION: There is provided the method in which a metabolic inhibitor of the enzymes or a manipulated source organism is used to diminish or avoid the action of the enzymes responsible for undesirable amino acids production or depletion of in vitro protein synthesis reactions. There is provided the method in which required high energy phosphate bonds are synthesized and supplied in situ so as to be synthesized, e.g. through coupling with an oxidation reaction as a homeostatic system for production of ATP. There is provided the method, in the homeostatic system, an energy source is supplied in combination with an enzyme that catalyzes the creation of the high energy phosphate bonds and with an enzyme that can use the high energy phosphate bonds to regenerate the ATP.
