PROBLEM TO BE SOLVED: To provide a method for examining a difference of base sequences of nucleic acid, with which whether the base sequence of nucleic acid to be a detection target has a difference from that of a nucleic acid to be a reference or not is examined.;SOLUTION: Firstly, a specimen containing a double-stranded DNAβ to be a reference and a double-stranded DNAβx to be a detection target is prepared. A double-stranded DNAβ containing a first double-stranded DNAβ1 provided with a first marker M1 and a second double-stranded DNAβ2 provided with a second marker M2 is prepared as the double-stranded DNAβ to be a reference. The second marker M2 has a property of changing optical properties when the second marker approaches the first marker M1. Then the double-stranded DNAs in the specimen are denatured. After that, the single-stranded DNAγ"s are reassociated. Then, optical properties of the specimen are examined during and/or after reassociation. Consequently, difference between the double-stranded DNAβ to be a reference and the double-stranded DNAβx to be a detection target can be examined.;COPYRIGHT: (C)2002,JPO
