Aspects of the invention provide methods for differential regulation of fatty acid unsaturation in seed oil and membrane lipids of plants, based on modulation of a previously unknown biosynthetic pathway involving a new phosphatidylcholine: diacylglycerol holinfosfotransferazu (PDCT), which regulates phosphatidylcholine biosynthesis in developing oilseed plants. Specific aspects relate to having the characteristic of the invention of the PDCT polypeptides, including, for example, variants, deletions, muteins, fusion proteins and their orthologs (collectively PDCT proteins), nucleic acids encoding them, plants containing such sequences or PDCT proteins, and to methods for generating plants having altered expression and / or activity of PDCT, including, but not limited to, methods including mutagenesis, recombinant DNA technology, transgenic plants, etc.
