DIGITAL IMAGING OF SINGLE RADIAL IMMUNODIFFUSION ASSAYS

A method for performing a single radial immunodiffusion (SRID) assay for anantigen or antibody of interest, comprising the steps of: (a) applying one ormore test sample(s) to a gel, wherein the gel contains (i) antibody specificto the antigen of interest and/or (ii) antigen recognised by the antibody ofinterest; (b) allowing the sample(s) to diffuse radially from its/theirpoint(s) of application; (c) preparing a digital image of the gel afterdiffusion has taken place; (d) determining the external diameter(s) ofprecipitation halos within the digital image; (e) comparing the determineddiameters to those resulting from one or more standard sample(s) containing aknown concentration of the antigen or antibody of interest; and, optionally,(f) using the results from step (e) to determine the concentration(s) of theantigen or antibody of interest in the test sample(s).
