The method involves determining kinetic clearances from measured kinetics, where the determined kinetic clearances are different from one another. A kinetic model is defined by a defining unit (20), where parameters of the defined model are formed by parameters of an input function and pharmacokinetic parameters. The input function and the pharmacokinetic parameters are estimated by resolution of the defined model from the determined kinetic clearances. Final input function and final pharmacokinetic parameters are deduced from the estimated input function and the pharmacokinetic parameters. Independent claims are also included for the following: (1) an information storage medium comprising a set of instructions for implementing a method for simultaneous extraction of input function and pharmacokinetic parameters of an active substance (2) a device for simultaneous extraction of input function and pharmacokinetic parameters of an active substance.
