PURPOSE: A method for preparing monosaccharide intestine of cuttlefish and chub mackerel is provided to effective perform pretreatment. CONSTITUTION: A method for preparing monosaccharide from intestine of cuttlefish and chub mackerel by supercritical treatment comprises: a step of removing water from the intestine and drying; a step of inputting dried ingredients, 60-99 weight% of extraction solvent of supercritical carbon dioxide, and 1-40 weight% of an aid solvent of ethanol, methanol, isopropanol, normal hexane, or water into an extraction column of 10C-100C and 40bar-500bar; a step of isolating fish oil ingredient, fish flavor ingredient, extraction solvent, and aid solvent and collecting only concentrated fish meat; a step of mixing the fish meat with catalyst and water; a step of pressurizing the mixture and hydrolyzing; and a step of cooling the hydrolyzed target material.
