PURPOSE: A method for manufacturing biofuel using microorganism and primer and probe use for the same are provided to qualify microorganism in real time and to stably produce biofuel. CONSTITUTION: A method for producing biofuel from biomass by microorganism fermentation comprises: a step of collecting a sample in which microorganism is fermenting; a step of qualifying degeneration of the microorganism in the sample; and a step of performing real time PCR. The microorganism is Clostridia sp. The Clostridia sp. microorganism is Clostridium acetobutylicum. The biofuel is butanol.
