The utilization of the substrate using a reaction adhesive i.e. cyanoacrylate, that penetrates and rapidly hardens epidermal barriers of skin. The substrate is applied on a skin surface and is removed after from the skin surface after the reaction adhesive is hardened. The substrate with the reaction adhesive and removed skin layers is utilized as a sample for determination of the epidermal barriers by an electron microscope or a scanning force microscope, where viscosity of the adhesive amounts to 70 to 100 centipoises at 25 degrees Celsius. Independent claims are also included for the following: (1) a method for realizing skin rupture (2) a method for non-invasive determination of epidermal barrier of skin (3) a device for non-invasive determination of epidermal barrier of skin.
