PROBLEM TO BE SOLVED: To provide a method that can accurately and efficiently quantitate cholesterol in a specific fraction by simple operation fundamentally without requiring polyanion, etc. and appropriately applicable to various automatic analyzers.SOLUTION: The method of pretreatment of a sample for quantitating cholesterol is characterized by affecting the sample containing lipoprotein with enzymes whose substrate is free cholesterol and if necessary, a reaction accelerator before measuring cholesterol present in a specific lipoprotein in the sample. There is also provided a method for quantitating cholesterol in the specific lipoprotein using the method of pretreatment, and a kit for quantitating cholesterol in the specific lipoprotein used in the method for quantification.
