PPROBLEM TO BE SOLVED: To provide a method of manufacturing a proton exchange membrane having a methanol suppression property and a high selectivity coefficient. PSOLUTION: In this method of manufacturing a proton exchange membrane, an unionized precursor of a proton exchange membrane is loosely irradiated with a gamma ray of 0.1-50 K gray (KGy); the precursor after the irradiation is formed into a proton exchange membrane by an ionization process of alkaline cleaning, acid cleaning and water cleaning. The proton exchange membrane has a proton conduction function, is improved in a methanol suppression property at the same time, can provide a further excellent relative selectivity coefficient, can be manufactured by a simple process because it is not necessary to unnecessarily add other raw materials and can be used as a barrier membrane of a methanol direct type fuel cell (DMFC) or a fuel cell of H01M008/10.
