Mutant p53 library

PPROBLEM TO BE SOLVED: To construct a comprehensive mutant gene library where at least one base is mutated in order to analyze p53 gene in view of the fact that it has been known that there are many mutations in the bases in the p53 gene as a cancer-restraining gene and it has been indicated that these mutations are closely correlated with the function to be fulfilled by the p53 gene. PSOLUTION: In order to efficiently construct the comprehensive mutant gene library, a 1st PCR is carried out using an oligonucleotide specifying mutation induction as a primer, and a 2nd PCR is then carried out using the PCR product from the 1st PCR as a megaprimer, and a gap repair vector is used in the PCR product thus obtained. This comprehensive mutant gene library thus obtained is useful not only in preparing nucleic acid arrays but also e.g. in analyzing the p53 gene because of being able to be utilized in the proteomics technology.
