METHOD TO SELECT EXPLANTS AND TO OBTAIN AN ASEPTIC CULTURE OF SMALL-LEAVED LIME (TILIA CORDATA MILL.)

A method to select explants and to obtain an aseptic culture of small-leaved lime (Tilia cordata Mill.) comprises obtaining the aseptic culture using explants of apices and segments of shoots of seedlings of the age from 2 months up to 1 year and sterilizing in a 0.1-0.3 % solution of diacide in the course of 15-25 min. Selection of vegetable material of 40-year trees in March-May, size and type of an explant, concentration and exposition of sterilizing substances: 70 % ethyl alcohol with exposure of 1 min., a 1 % AgNOsolution 15 min. and 2.5 % NaClO - 10 min., and composition of a nutrient medium according to the Murashige and Skoog prescription adding activated coal in a concentration of 1-2 g∙l.
