PURPOSE: A DNA chip using a random Listeria monocytogenes DNA fragment is provided to detect only Listeria monocytogenes. CONSTITUTION: A DNA chip for detecting Listeria monocytogenes contains a genomic DNA-derived random Listeria monocytogenes genomic DNA probe. The DNA probe is a 100-1500 bp DNA prepared from genomic DNA isolated from Listeria monocytogenes by cleavage with different restriction enzyme pairs. The restriction enzyme pair includes EcoR I/BamH I, Hind III/Xho I and HindIII/Sac II. A method for manufacturing the DNA chip comprises: a step of isolating genomic DNA from Listeria monocytogenes; a step of cleaving the genomic DNA by different restriction enzyme pairs; a step of performing a ligation of the cleaved genomic DNA with a plasmid for library to obtain a colony; a step of isolating plasmid from the colony and performing PCR with T3/T7 primer; and a step of fixing the probe on a slide glass.
