Compositions and Methods for direct Protein antigens to dc through the Human receptor asialoglicoproteu00ednas DC (DC ASGPR). The dc ASGPR presents an intracellular tyrosine based motif and dileucina which results in the generation of Regulatory T cells and il-10 in vitro and in vivo.The present methods can be used to design vaccines against those with defined autoantigens in autoimmune diseases such as type 1 Diabetes and multiple sclerosis.Claim 1: a method to generate one or more Regulatory T Cells (Treg) with specificity for an antigen, which comprises Isolating one or more Human dendritic cells (DC) of a subject; to load one or more antigens in one or more dc with a specific Antibody against the receiver Of the asialoglicoproteu00ednas (DC - DC ASGPR), or a fragment of this Union,Conjugated or fused with one or more antigens to form dc loaded with the Antigen; and contacting the DC loaded with the Antigen with one or more non stimulated T cells, where the DC loaded with the Antigen stimulated the Proliferation of Treg with specificity for the Antigen.
